Casey Mittelstadt had one goal and one assist and Owen Power also scored for the Sabres. Alex Tuch contributed two assists.
Sabres goaltender Ukko-Pekka Luukkonen made 38 saves.
Joel Farabee and Cam Atkinson scored one goal apiece for the Flyers, who have dropped three games in a row.
Travis Sanheim had two assists.
Flyers goaltender Carter Hart made three saves on five shots in the opening 10 minutes before being replaced by Samuel Ersson due to a mid-body injury. Ersson finished with seven saves.
The Sabres took a 3-2 lead at 3:03 of the third period thanks to a laser of a wrist shot by Power that clanged off the inside of the post and into the net.
Philadelphia's Tyson Foerster ripped a slap shot at 9:25, but the puck was deflected into the stands.
After Luukkonen made several stellar saves, the Sabres added to their lead as Thompson connected at 13:28 for a 4-2 advantage.
Ersson was pulled for an extra skater at 16:08 and Biro scored into the empty net at 16:52 for a 5-2 lead.
The Flyers went ahead 55 seconds into the first when Farabee was credited with a goal after the puck appeared to deflect off Buffalo defenseman Henri Jokiharju.
The Sabres equalized at 3:38 when Mittelstadt scored on his team's first shot.
Buffalo raced out on a five-on-three break and appeared to take a one-goal advantage at 9:29 when Jeff Skinner scored. The Flyers utilized their coach's challenge for offside, and the goal was disallowed. Just 31 seconds later, Biro notched his first career goal.
Philadelphia tied things at 16:17 when Atkinson redirected Sanheim's pass into the net.
The Sabres controlled the tempo for much of the second, and the aggressive play paid off as they were awarded a power play at 18:27 when Nicolas Deslauriers was sent off for roughing. The period ended, however, without Buffalo finding the back of the net.
